Introduction
Why study microbes?
- Microbes have existed on earth for over 3 billion years
- Prokaryotes genetically are far more diverse than eukaryotes
- Life on this planet as we know it would not have existed without microbes
- Less than 1% of microbial diversity that exists on earth has been cultured
- Microbes make up the single most important source for the biotechnology industry

Given the importance of microbes on our planet (and perhaps on other planets e.g. Mars) and their roles in infectious diseases, the identification and characterisation has become enormously important. Modern, effective and complete identification and characterisation of microbes requires that one is conversant in both culturing and molecular techniques. This course enables students to identify and differentiate microbes using conventional culturing techniques, microscopy and modern molecular / phylogenetic approaches. Currently the single most important technique in microbial identification and characterisation is 16S rRNA, consequently it will be a major focus of the course.

Project aim
Perform a range of phenotypic and phylogenetic experiments on an Escherichia coli strain of unknown origin. From the results of the phylogenetic microbial source tracking (MST) experiments determine the possible source of the strain (Human or animal). Consolidate the results of the phenotypic and phylogenetic experiments with the published literature (to date) on Escherichia coli, microbial source tracking (MST), phylogenetic and phenotypic characterisation formulate a discussion and conclusion. Write up your methodology, results, discussion and conclusion into a report in the style of a microbial research paper (or journal article). Accompany the written report with a short technical seminar where you will present your experimental results and findings to the class.
