Abstract A

Identification of unknown pathogenic bacteria is critical for correct diagnosis of diseases and effective prevention against the spread of foodborne pathogens in commercially packaged produce. This study was undertaken to objectively assess traditional biochemical tests and the more recent 16s rRNA molecular sequencing method for bacterial identification of an unknown mixed culture. From an analysis of biochemical test results alonci the prediction of the bacterial colony 1 could only be narrowed down to either Citrobocter freundii, Proteus mirabilis or Proteus vulgaris. Critically important biochemical test results contradicted those outlined in first stage test tables for P. vulgaris so an accurate prediction of this bacteria could not be made. Conversely, 16s rRNA sequencing of the bacterial colony 2 elicited clear and accurate sequencing results so a precise prediction of Enterococcus faecalis was made for this bacteria. Against this background, molecular sequencing was considered far more reliable and accurate than traditional biochemical assays. 
 

Abstract B

The aim of this experiment is to use both biochemical and bimolecular test techniques and compare to see which set of tests is more accurate and gives better results. To do this we had to identify and isolate 3 unknown bacterial samples. The samples we had were Proteus mirabilis. Micrococcus luteus and. Staphylococcus epidermidis. However, the answers did not always coincide with our results due to many different factors, both environmental and simple human error. With the gene sequencing test BLAST which is a program that analyses the proteins and contaminants in your isolated bacterial sample the results showed a 98% match to both Proteus mirabilis and Proteus vulgaris (Seen in Fig 1). We then had to decide which one it was most likely to be based on the simple biological tests however we came up with the wrong one. The other two bacterial samples were not conclusive with our tests and this is most likely due to contamination of our sample or human error in the experimental processes.
