The major piece of work for this module is a scientific research article. A multiplex PCR has been developed to characterize strains of Vibrio cholerae. The reaction amplifies four different target genes. You have been provided with the agarose gel results after electrophoresis (see file ‘Agarose Gel Results.jpg’). Your report will provide background to currently used diagnostic assays, and analyse the results from this gel.

Introduction (approximately 1000 words)

Explain how advances in molecular microbiology are being used to develop new diagnostic assays. PCR is currently used as a diagnostic method – in addition to discussing the advantages of PCR, name and describe two other methods (not including alternative forms of PCR), and for each one, provide an example of a diagnostic assay it is used for (e.g. for a particular species of type of infection).

Results (approximately 500 words)

A multiplex PCR has been developed to characterize strains of Vibrio cholerae. The reaction amplifies four different target genes:

1. The largest, a core gene (piuB) which should be present in all strains.
2. Two genes (ctxA and ctxB), which are present in some strains, but not in others. The same size fragment, of medium size, is amplified for these two genes.
3. The smallest, gene norM, which is also variably present.

Using the gel image provided (see file ‘Agarose Gel Results.jpg’), indicate on the gel which of the bands is gene piuB, which bands are genes ctxA and ctxB, and which band is gene norM. Using the ladder on the gel (lane 8), and the ladder provided in file ‘Ladder.jpg’, estimate the size of each of these bands.

Seven strains have been tested (lanes 1-7). State what genes each of the seven strains are positive for.

Discussion (approximately 1000 words).

Critically evaluate the multiplex PCR (what is good about it, what is bad about it. Explain why).

Explain why a researcher would want to include these four genes in a multiplex assay.
