Method evaluation
St. Hatchcroft Hospital Clinical Biochemistry department is undertaking an evaluation of a new rapid immunological method for the measurement of oestradiol that can determine results in just a few minutes. Although the old method is extremely accurate, it takes many hours to complete. They are assessing the new method in terms of precision accuracy (comparison with old method) and cross-reactivity.
1. The data in sheet 1 shows the results of repeated measurements of standard oestradiol samples of known concentration. Use this information to create a plot of concentration against the coefficient of variation (precision profile). Comment on what the results of this profile tells you about the useful range of this assay.
2. The data in sheet 2 shows the result of a comparison between this new method and the method used previously. Undertake a comparison of these methods by carrying out a Bland Altman plot. What do the results of tell you about the relationship between these two methods and the usefulness of this new assay?
3. The data in the sheet shows the absorbance values obtained when known concentrations of estrone are used instead of oestradiol in this assay. Use this data to calculate the % cross-reactivity of oestrone.
You can draw the plots in any way that you like - graph paper, excel, etc. However, make sure that these plots are easy to interpret and carry the correct information. When doing calculations, please show how you have derived your answers (show working out).
