Excised EDL and SOL muscles were placed into individual organ baths (Danish Myogenic Technology, Denmark) filled with Krebs-Henseleit Ringer’s solution (containing in millimoles per litre: NaCl 118, KCl 4.75, MgSO47H2O 1.18, Na2HPO4 1, CaCl2H2O 2.5, NaHCO3 24 and glucose 11). Each organ bath was bubbled with carbogen (5% CO₂ in O₂) and maintained at a temperature of 30°C and a pH of 7.4. Data was collected and analysed using LabChart Pro version 8.0 software, customized for this experiment (ADInstruments, NZ). At proximal end, the muscle was hooked (via knotted surgical silk loops) onto a previously calibrated force transducer and the distal end was fixed to a micromanipulator with stimulating electrodes flanking the muscle belly. Before the commencement of the contractility experiments, optimal length (L_o) for each muscle was determined to ensure maximal contractile force generation. L_o for each muscle was obtained via a series of twitch contractions at increasing lengths thereby ensuring optimal overlap of the sarcomeres of each muscle. Supramaximal square wave pulses (100 Hz, 0.2ms duration) were delivered to muscles at a voltage of 21V for the EDL and SOL in order to produce maximum isometric tetanic contractions (P_o). Determination of L_o was measured with callipers and once established for both muscles, a twitch response was administered followed by a single tetanic stimulation at 100 Hz.
