You are asked to watch a total of seven short YouTube videos relating to plating, counting, and identifying bacteria, and looking at the effects of antibiotics. Please watch the videos in the order they are listed here, and answer any questions associated with each procedure.
Video 1. In video 1 (thank you Lila, Mo, and Elliot) you are introduced to the method of making serial dilutions, which will be used to assess bacterial counts in three probiotic yoghurts.
In Video 2 (also in Weblearn), you are led through (i) the methods of determining the original bacterial count (cfu) in the yogurt, (ii) ways in which to assess the morphology of different cultures, and (iii) how to carry out the Gram staining procedure.

2. If I make three sequential 1:10 serial dilutions of a suspension of bacteria originally containing 1.00 x10 7 cfu/mL, how many cfu/mL would my final suspension contain? (show your workings).
3. In Table 1 below, the number of colonies counted for each yogurt, at each of three serial dilutions, are shown.
a) Complete the right-had column of the table by calculating the cfu per gram of yoghurt, using the method shown in video 2. (Remember, you should only do this where the colony counts are fall within a particular range per plate – this range is stated in the video; where this is not the case, simply write n/a).
4. Gram staining is named after Danish bacteriologist Hans Christian Gram, who developed the technique. Watch the Khan Academy video below to find out more about this, and also about the different shapes of bacteria. Video 3: Bacterial characteristics – Gram staining.
